The mechanism by which the linker mediates the concerted action of the two domains remains a conundrum. Our SAXS data shows that ThCel7A linker is partially-extended in solution. Structural modeling suggests that this linker conformation is stabilized by inter- and intra-molecular interactions involving the linker peptide and its O-glycosylations.

The peroxisome proliferator-activated receptors (PPARs) regulate genes involved in lipid and carbohydrate metabolism, and are targets of drugs approved for human use. Whereas the crystallographic structure of the complex of full length PPARγ and RXRα is known, structural alterations induced by heterodimer formation and DNA contacts are not well understood. Herein, we report a small-angle X-ray scattering analysis of the oligomeric state of hPPARγ alone and in the presence of retinoid X receptor (RXR). The results reveal that, in contrast with other studied nuclear receptors, which predominantly form dimers in solution, hPPARγ remains in the monomeric form by itself but forms heterodimers with hRXRα.
Understanding the adsorption mechanisms in nanostructured polymer films has become crucial for their use in technological applications, since film properties vary considerably with the experimental conditions utilized for film fabrication. In this paper, we employ small-angle X-ray scattering (SAXS) to investigate solutions of polyanilines and correlate the chain conformations with morphological features of the nanostructured films obtained with atomic force microscopy (AFM). It is shown that aggregates formed already in solution affect the film morphology; in particular, at early stages of adsorption film morphology appears entirely governed by the chain conformation in solution and adsorption of aggregates.
